Methyl 4-hydroxybenzoate, also methyl paraben, one of the parabens, is a preservative with the chemical formula CH3(C6H4(OH)COO). It is the methyl ester of p-hydroxybenzoic acid. Methyl 4-hydroxybenzoate is a 4-hydroxybenzoate ester resulting from the formal condensation of the carboxy group of 4-hydroxybenzoic acid with methanol. It is the most frequently used antimicrobial preservative in food, medicine and cosmetics. It occurs naturally in several fruits, particularly in blueberries. It has a role as a plant metabolite, an antimicrobial food preservative, a neuroprotective agent and an antifungal agent. Methyl 4-hydroxybenzoate is a Standardized Chemical Allergen. The physiologic effect of methylparaben is by means of Increased Histamine Release, and Cell-mediated Immunity.
Methyl 4-hydroxybenzoate is obtained by esterification of p-hydroxybenzoic acid with methanol. Para-hydroxybenzoic acid was added to excess methanol to dissolve, and concentrated sulfuric acid was slowly added with stirring. After heating under reflux for 10h, poured into water to precipitate crystals, washed with water and sodium carbonate solution to obtain crude product. The finished product is obtained by recrystallization with water or 25% ethanol. The yield was 85%. Raw material consumption (kg/t) p-hydroxybenzoic acid 1200 methanol 1000.
Methyl 4-hydroxybenzoate has the following applications/functions:
1. Methyl 4-hydroxybenzoate may be used as a pharmaceutical reference standard for the quantification of the analyte in pharmaceutical formulations using reversed-phase sequential injection chromatography (SIC) technique and high-performance liquid chromatography technique.
2. Methyl 4-hydroxybenzoate is an anti-fungal agent often used in a variety of cosmetics and personal-care products. It is also used as a food preservative. It can also be used in medicine and organic synthesis.
3. Methyl 4-hydroxybenzoate is commonly used as a fungicide in Drosophila food media at 0.1%. To Drosophila, methylparaben is toxic at higher concentrations, has an estrogenic effect (mimicking estrogen in rats and having anti-androgenic activity), and slows the growth rate in the larval and pupal stages at 0.2%.
4. Methyl 4-hydroxybenzoate can also be used as an antimicrobial agent. It is used in the study of steroid hormone synthesis.
