Polymyxin-B-sulfate is a member of the polymyxin group of antibiotics, obtained from different strains of Bacillus polymyxa and related species. It contains a cyclic heptapeptide unit and a tripeptide side chain, which is N-acylated by a fatty acyl residue. Polymyxin-B-sulfate is one of a group of basic polypeptide antibiotics derived from B polymyxa (B aerosporous). Polymyxin-B-sulfate is the sulfate salt of Polymyxins B1 and B2, which are produced by the growth of Bacillus polymyxa (Prazmowski) Migula (Fam. Bacillacea). It has a potency of not less than 6000 polymyxin B (polymyxin b sulfate) units per mg, calculated on the anhydrous basis. Each vial contains 500,000 polymyxin B (polymyxin b sulfate) units for parenteral or ophthalmic administration. Polymyxin B (polymyxin b sulfate) forInjection is in powder form suitable for preparation of sterile solutions for intramuscular, intravenous drip, intrathecal, or ophthalmic use.
Aqueous solutions of polymyxin B sulfate may be stored up to 12 months without significant loss of potency if kept under refrigeration.In the interest of safety, solutions for parenteral use should be stored under refrigeration and any unused portion should be discarded after 72 hours. Polymyxin-B-sulfate should not be stored in alkaline solutions since they are less stable.
Polymyxin-B-sulfate has the following applications/functions:
1. Polymyxin-B-sulfate is similar to polymyxin E, and has inhibitory effect on gram-negative bacilli, such as Escherichia coli, Pseudomonas aeruginosa, paraEscherichia coli, Klebsiella pneumoniae, acidophilus, pertussis bacillus and dysentery bacillus or bactericidal effect.
2. Polymyxin-B-sulfate is mainly used for infection of wounds, urinary tract, eyes, ears, trachea and other parts caused by Pseudomonas aeruginosa and other pseudomonas. It can also be used for sepsis and peritonitis.
3. Polymyxin-B-sulfate can also be used for serious infections caused by aminoglycoside-resistant, third-generation cephalosporin-resistant bacteria and Pseudomonas aeruginosa or other susceptible bacteria, such as bacteremia, endocarditis, pneumonia, post-burn infection, etc.
