Semaglutide is an anti-diabetic medicatin used for the treatment of type 2 diabetes and chronic weight management. Semaglutide acts like human glucagon-like peptide-1 (GLP-1) such that it increases insulin secretion, thereby increasing sugar metabolism. It is distributed as a metered subcutaneous injection in a prefilled pen, or as an oral form. One of its advantages over other antidiabetic drugs is that it has a long duration of action, thus, only once-a-week injection is sufficient.

1、Semaglutide is a GLP-1 analog with 94% sequence homology with human GLP-1. Semaglutide acts as a GLP-1 receptor agonist, which selectively binds to and activates the GLP-1 receptor (the target of natural GLP-1). The main mechanism leading to the prolonged half-life of semaglutide is albumin binding, which leads to reduced renal clearance and prevents metabolic degradation. In addition, simaglutide is stable against degradation by the DPP-4 enzyme. Semaglutide lowers blood sugar by stimulating insulin secretion and reducing glucagon secretion mechanisms, both of which occur in a glucose-dependent manner. Therefore, when blood sugar is high, insulin secretion is stimulated and glucagon secretion is inhibited. The mechanism of blood sugar reduction also involves a slight delay in gastric emptying in the early postprandial period.
2、In the United States, semaglutide is also indicated as an adjunct to diet and exercise for chronic weight management in adults with obesity (initial body mass index (BMI) ≥ 30 kg/m2) or overweight (initial BMI ≥ 27 kg/m2) with at least one weight-related comorbidity.Semaglutide is a glucagon-like peptide-1 receptor agonist.It reduces food intake by lowering appetite and slows down digestion in the stomach.[25] In this way it reduces body fat.
